Speaking On:Genomic Analysis of Hirschsprung Disease
It is commonly assumed that only very large numbers of cases and extensive sequencing is necessary for uncovering the genetic basis of complex disease. Although this is undoubtedly true for some (many?) traits and diseases one cannot ignore the possibility that complexity in INHERITANCE can arise from even a few genes. Sentinel examples in human genetics have frequently illustrated the mechanisms of disease that have then been more broadly observed. I will advance the notion that, as in early molecular biology, and human genetics, intense study of a few examples to understand disease mechanisms in complex inheritance may be equally worthwhile. I will describe our studies of Hirschsprung disease as one such model genomic disorder.
